Synthetic negative- and positive-strand RNA templates were produced by T7 RNA polymerase and serial dilutions were prepared in total cellular RNA (from 2x105 CESS cells) and tested by strand-specific RTPCR assay. After electrophoresis, the bands were visualized by ethidium bromide staining. Specificity was confirmed by testing with or without the reverse transcription step.
